Development of methods effective for detection and decrease of host-cell proteins from therapeutical antibodies
Targets of the project:
- Development of a new, very sensitive and selective method, applicable for high throughput applications, to quantify host cell proteins (HCP). Furthermore this method should facilitate the decrease of particular HCPs in a pool of HCPs even in the presence of high concentrations of active agents. During the several chromatography projects the decreasing rate should be detected quantitatively and with highest sensitivity.
- Identification of those HCP, which are difficult to remove in antibody-isolation processes.
- Solution for efficient decrease of these selected HCP in column chromatography based on their specific protein-chemical characteristics and accompanied with mp-MS-quant Analytic.
Conclusion: In case of success, bottle necks in downstream processing during the production of bio-pharmaceutical proteins will be abandoned and in addition the whole process will be much saver and will fulfill all quality standards. Furthermore the production activity will become more economical.
